Combination therapywithtwoormoreanalgesicsiswidelyusedforconditionsassociatedwithmod-
eratetoseverepain.Combinationsofdiverseanalgesicswithdifferentmodesofactioncanimprovethe
risk-benefit ratioofanalgesictreatments.Theaimofthisstudyistoevaluatetheantinociceptiveeffectof
tapentadol (TAP)and flupirtine (FLP),whenadministeredseparatelyorincombination,aswellastheir
synergistic interactionintheorofacialformalintestinrats.Afteri.p.injectionofTAPatdifferentdoses(2,
5, 10and15mg/kg),thebiphasicnociceptivebehaviorwasreducedinadose-dependentmannerinboth
phase IandII.Conversely,i.p.injectionofFLPatdifferentdoses(0.6,1.6,3.3,6.6,16.6and22.2mg/kg)
induced adose-dependentantinociceptiveeffectinphaseIIonly.TAPwasfoundtobemoreeffective
than FLP.TheinteractionbetweenTAPandFLPwassynergisticinphaseIIwithaninteractionindex(γ) of
0.5070.24. ThedatareportedinthisstudyindicatethatFLPenhancestheantinociceptiveeffectofTAP
and thisdrugcombinationmightbepotentiallyusefulinthetreatmentofchronicpain
